Homopolymeric region followed by a non-recognizable sequence

Polymerase slippage - polyT - raw-website

Polymerase slippage - polyT - electropherogram-website

Long homopolymeric region (usually polyT or polyA) followed by overlapping peaks

Possible causes

Slippage of the DNA polymerase off the template after a repeat region

Sequence the complementary strand.

Design a new primer to hybridize just outside the repeat region.

If a PCR product, clone it. Polymerase slippage after a homopolymeric region is more pronounced in PCR products than plasmid prep.
